Questions-PhyTip Columns
2.5. Analytical methods for cell culture
Cell counts and viability measurements were performed using a Vi- Cell analyzer (Beckmann Coulter, Brea, CA) and titers were quantified using a protein affinity high performance liquid chromatography device (PA-HPLC, Waters, Baden-Wättwil, Switzerland). Protein quality of Phytips eluates (PhytipsVR, PhyNexus, San Jose, California, USA) were analyzed by imaged capillary isoelectric focusing (iCE280 analyzer, ProteinSimple, Santa Clara, CA) for charge variants, by capillary gel electrophoresis with laser-induced fluorescence detection (CGE-LIF, DNA genetic analyzer 3130XL, Life Technologies, Darmstadt, Germany) for glycosylation, by reduced caliper (Caliper LabChip GXII, Perkin Elmer, Waltham, Massachusetts, USA) for the determination of clipping and by size exclusion high performance liquid chromatography (SE- HPLC, Waters, Baden-Wättwil, Switzerland) for aggregates.
